PT00114 and PT00121 are peptide-based formulations currently in preclinical development for the treatment of anxiety and depression disorders.
PT00114
PT00114 is a peptide formulation based on a new class of compounds with a unique mode of action and is currently in preclinical development for the treatment of anxiety and depression disorders. This product is based on a recently discovered peptide that protects cells from stress and promotes communication among brain cells. PT00114 is unusual among peptides in that it selectively targets cells under stress and does not have an impact on normal unstressed cells. The underlying mechanism of PT00114’s action is its ability to rescue neuronal cells from physiological insult.
PT00114 is efficacious in animal models at low doses, has a rapid onset and a long duration of action. It acts, in part, by inhibiting the effects of corticotropin-releasing hormone (CRH), the main stress inducing hormone in the body. In animals, it crosses the blood-brain barrier and inhibits the effects of high stress and anxiety but has little effect in unstressed animals. Thus, PT00114 represents true stress-effect targeted therapy that is applicable to a wide range of mood disorders. This suggests that as a treatment, PT00114 has the potential to inhibit the harmful effects of stress on the patient without causing unwanted side effects.
The neuroprotective capability of PT00114 formulations is the basis of its use in spinal cord injuries, trauma, ischemia and neurodegenerative disorders that are currently under investigation.
PT00121
PT00121 is a newer formulation incorporating a shorter peptide as the active component and shows many of the same functional properties as PT00114 in vitro. It is currently being tested in vivo.
